研究室紹介

Professor Sung-Hyun Kim's research lab focuses on cellular and molecular mechanisms underlying neural development, synaptic function, and vascular integrity in disease. The lab investigates key regulators of cell adhesion and morphogenesis during vertebrate gastrulation, synaptic vesicle trafficking and neurotransmitter release, and endothelial protection in sepsis. A central theme is the molecular machinery controlling cell polarity, membrane dynamics, and signaling pathways in both neurological and vascular pathologies. The lab integrates molecular biology, live imaging, and in vivo models to dissect fundamental processes in development and disease.

The role of Paraxial Protocadherin in selective adhesion and cell movements of the mesoderm during Xenopus gastrulation
Sung‐Hyun Kim, Akihito Yamamoto, Tewis Bouwmeester, Eric Agius, Edward M. De Robertis
SJR Q1Development

Paraxial Protocadherin (PAPC) encodes a transmembrane protein expressed initially in Spemann's organizer and then in paraxial mesoderm. Together with another member of the protocadherin family, Axial Protocadherin (AXPC), it subdivides gastrulating mesoderm into paraxial and axial domains. PAPC has potent homotypic cell adhesion activity in cell dissociation and reaggregation assays. Gain- and loss-of-function microinjection studies indicate that PAPC plays an important role in the convergence a

Protection of endothelial integrity has been recognized as a frontline approach to alleviating sepsis progression, yet no effective agent for preserving endothelial integrity is available. Using an unusual anti-angiopoietin 2 (ANG2) antibody, ABTAA (ANG2-binding and TIE2-activating antibody), we show that activation of the endothelial receptor TIE2 protects the vasculature from septic damage and provides survival benefit in three sepsis mouse models. Upon binding to ANG2, ABTAA triggers clusteri

Synaptic Vesicle Recycling at CNS Synapses without AP-2
Sung Hyun Kim, Timothy A. Ryan
SJR Q1Journal of NeuroscienceOA

Synaptic vesicles (SVs) are composed of approximately 10 types of transmembrane proteins that must be recycled after exocytosis of neurotransmitter. The mechanisms for resorting these proteins into synaptic vesicles once incorporated into the plasma membrane after exocytosis are poorly understood. The adaptor complex AP-2 is the major clathrin-associated adaptor for cargo recognition at the plasma membrane. Here, we have investigated its role in synaptic vesicle endocytosis. shRNA-mediated knock

Synapses in neurodegenerative diseases
Jae Ryul Bae, Sung Hyun Kim
SJR Q1BMB ReportsOA

Synapse is the basic structural and functional component for neural communication in the brain. The presynaptic terminal is the structural and functionally essential area that initiates communication and maintains the continuous functional neural information flow. It contains synaptic vesicles (SV) filled with neurotransmitters, an active zone for release, and numerous proteins for SV fusion and retrieval. The structural and functional synaptic plasticity is a representative characteristic; howe

Balance of Calcineurin Aα and CDK5 Activities Sets Release Probability at Nerve Terminals
Sung Hyun Kim, Timothy A. Ryan
SJR Q1Journal of NeuroscienceOA

The control of neurotransmitter release at nerve terminals is of profound importance for neurological function and provides a powerful control system in neural networks. We show that the balance of enzymatic activities of the α isoform of the phosphatase calcineurin (CNAα) and the kinase cyclin-dependent kinase 5 (CDK5) has a dramatic influence over single action potential (AP)-driven exocytosis at nerve terminals. Acute or chronic loss of these enzymatic activities results in a sevenfold impact

Long non-coding RNAs in brain tumors: roles and potential as therapeutic targets
Sung‐Hyun Kim, Key‐Hwan Lim, Sumin Yang, Jae‐Yeol Joo
SJR Q1Journal of Hematology & OncologyOA

Brain tumors are associated with adverse outcomes despite improvements in radiation therapy, chemotherapy, and photodynamic therapy. However, treatment approaches are evolving, and new biological phenomena are being explored to identify the appropriate treatment of brain tumors. Long non-coding RNAs (lncRNAs), a type of non-coding RNA longer than 200 nucleotides, regulate gene expression at the transcriptional, post-transcriptional, and epigenetic levels and are involved in a variety of biologic

Distinct synaptic vesicle recycling in inhibitory nerve terminals is coordinated by SV2A
Jae Ryul Bae, Wongyoung Lee, Young Ok Jo, Sukmin Han, Soulmee Koh, Woo Keun Song, Sung Hyun Kim
SJR Q1Progress in NeurobiologyOA

Proper brain function requires a balance between excitatory and inhibitory neuronal activity. This balance, which is disrupted in various neural disorders, ultimately depends on the functional properties of both excitatory and inhibitory neurons; however, how the physiological properties of presynaptic terminals are controlled in these neurons is largely unknown. In this study, we generated pHluorin-conjugated, synaptic vesicle-specific tracers that are preferentially expressed in excitatory or

Idiopathic Synovial Osteochondromatosis of the Hip: Radiographic and MR Appearances in 15 Patients
Sung Hyun Kim, Suk Ju Hong, Ji Seon Park, Jae Min Cho, Eung Yeop Kim, Joong Mo Ahn, Youn Soo Park
SJR Q1Korean Journal of RadiologyOA

The most common radiographic finding of synovial osteochondromatosis of the hip was the presence of juxta-articular calcified and/ or ossified bodies. MR imaging depicted intra-articular bodies of focal low signal intensity at all pulse sequences, with areas of iso-intensity at T1WI and hyperintensity at T2WI. In addition, the presence of an extra-articular herniation sac was not uncommon.

Bioactive Adsorber Model for Industrial Wastewater Treatment
Sung‐Hyun Kim, Massoud Pirbazari
SJR Q3Journal of Environmental Engineering

A predictive mathematical model that describes the adsorption and biodegradation phenomena in recycle fluidized‐bed (RFB) adsorbers was developed. The model incorporated liquid film transfer, biodegradation and diffusion in the biofilm, adsorption onto activated carbon, and biofilm growth. The model equations were solved by a combinatorial technique involving the methods of orthogonal collocation and finite differences. Computer simulations of the model were used for adsorber performance predict
