Professor Yeonhee Park's research lab specializes in statistical methodology and its applications in biomedical and health sciences, with a strong focus on precision medicine, genetic association studies, and clinical trial design. The lab develops advanced statistical models—such as groupwise envelope models and Bayesian enrichment designs—to improve the efficiency and accuracy of detecting associations between genetic variants and complex phenotypes, particularly in neuroimaging and patient outcome data. A key emphasis is on addressing real-world challenges in clinical research, including non-normal data distributions, small sample sizes, and the need for robust, interpretable statistical inference. The lab also investigates financial reporting practices in financial institutions, applying statistical methods to detect earnings management and capital adequacy manipulation.

Altered Lipid Synthesis by Lack of Yeast Pah1 Phosphatidate Phosphatase Reduces Chronological Life Span
Yeonhee Park, Gil-Soo Han, Eugenia Mileykovskaya, Teresa A. Garrett, George Carman
SJR Q1Journal of Biological ChemistryOA

In Saccharomyces cerevisiae, Pah1 phosphatidate phosphatase, which catalyzes the dephosphorylation of phosphatidate to yield diacylglycerol, plays a crucial role in the synthesis of the storage lipid triacylglycerol. This evolutionarily conserved enzyme also plays a negative regulatory role in controlling de novo membrane phospholipid synthesis through its consumption of phosphatidate. We found that the pah1Δ mutant was defective in the utilization of non-fermentable carbon sources but not in ox

Bayesian group sequential enrichment designs based on adaptive regression of response and survival time on baseline biomarkers
Yeonhee Park, Suyu Liu, Peter F. Thall, Ying Yuan
SJR Q1BiometricsOA

Precision medicine relies on the idea that, for a particular targeted agent, only a subpopulation of patients is sensitive to it and thus may benefit from it therapeutically. In practice, it is often assumed based on preclinical data that a treatment-sensitive subpopulation is known, and moreover that the agent is substantively efficacious in that subpopulation. Due to important differences between preclinical settings and human biology, however, data from patients treated with a new targeted ag

Groupwise Envelope Models for Imaging Genetic Analysis
Yeonhee Park, Zhihua Su, Hongtu Zhu
SJR Q1Biometrics

Motivated by searching for associations between genetic variants and brain imaging phenotypes, the aim of this article is to develop a groupwise envelope model for multivariate linear regression in order to establish the association between both multivariate responses and covariates. The groupwise envelope model allows for both distinct regression coefficients and distinct error structures for different groups. Statistically, the proposed envelope model can dramatically improve efficiency of tes

Optimal two-stage group sequential designs based on Mann-Whitney-Wilcoxontest
Yeonhee Park
SJR Q1PLoS ONEOA

The Mann-Whitney-Wilcoxon test, often referred to as the Mann-Whitney U test or Wilcoxon rank-sum test, is a non-parametric statistical test used to compare two independent groups when the dependent variable is ordinal or continuous but not normally distributed. It's particularly useful for small sample sizes or when the assumptions of parametric tests, such as the t-test, are violated, including cases where the data is skewed. This study focuses on the Mann-Whitney-Wilcoxon test for ordinal dat

Envelope‐based partial partial least squares with application to cytokine‐based biomarker analysis for COVID‐19
Yeonhee Park, Zhihua Su, Dongjun Chung
SJR Q1Statistics in MedicineOA

Partial least squares (PLS) regression is a popular alternative to ordinary least squares regression because of its superior prediction performance demonstrated in many cases. In various contemporary applications, the predictors include both continuous and categorical variables. A common practice in PLS regression is to treat the categorical variable as continuous. However, studies find that this practice may lead to biased estimates and invalid inferences (Schuberth et al., 2018). Based on a co

Personalized Risk-Based Screening Design for Comparative Two-Arm Group Sequential Clinical Trials
Yeonhee Park
SJR Q2Journal of Personalized MedicineOA

Personalized medicine has been emerging to take into account individual variability in genes and environment. In the era of personalized medicine, it is critical to incorporate the patients' characteristics and improve the clinical benefit for patients. The patients' characteristics are incorporated in adaptive randomization to identify patients who are expected to get more benefit from the treatment and optimize the treatment allocation. However, it is challenging to control potential selection

환헤지 통화옵션 상품의 손실 현황과 대응방안: KIKO(Knock-In Knock-Out) 거래를 중심으로

Optimal two-stage design of single arm Phase II clinical trials based on median event time test
Yeonhee Park
SJR Q1PLoS ONEOA

The Phase II clinical trials aim to assess the therapeutic efficacy of a new drug. The therapeutic efficacy has been often quantified by response rate such as overall response rate or survival probability in the Phase II setting. However, there is a strong desire to use survival time, which is the gold standard endpoint for the confirmatory Phase III study, when investigators set the primary objective of the Phase II study and test hypotheses based on the median survivals. We propose a method fo
