The DNA damage response induces inflammation and senescence by inhibiting autophagy of GATA4

SJR Q1ScienceOA

Cellular senescence is a terminal stress-activated program controlled by the p53 and p16(INK4a) tumor suppressor proteins. A striking feature of senescence is the senescence-associated secretory phenotype (SASP), a pro-inflammatory response linked to tumor promotion and aging. We have identified the transcription factor GATA4 as a senescence and SASP regulator. GATA4 is stabilized in cells undergoing senescence and is required for the SASP. Normally, GATA4 is degraded by p62-mediated selective a

Dual roles of autophagy in the survival of Caenorhabditis elegans during starvation
Chanhee Kang, Young‐Jai You, Leon Avery
SJR Q1Genes & DevelopmentOA

Autophagy is a major pathway used to degrade long-lived proteins and organelles. Autophagy is thought to promote both cell and organism survival by providing fundamental building blocks to maintain energy homeostasis during starvation. Under different conditions, however, autophagy may instead act to promote cell death through an autophagic cell death pathway distinct from apoptosis. Although several recent papers suggest that autophagy plays a role in cell death, it is not known whether autopha

How autophagy both activates and inhibits cellular senescence
Chanhee Kang, Stephen J. Elledge
SJR Q1AutophagyOA

Autophagy and cellular senescence are stress responses essential for homeostasis. While recent studies indicate a genetic relationship between autophagy and senescence, whether autophagy acts positively or negatively on senescence is still subject to debate. Although autophagy was originally recognized as a nonspecific lysosomal degradation pathway (general autophagy), increasing evidence supports a selective form of autophagy that mediates the degradation of specific targets (selective autophag

Autophagy Is Pro-Senescence When Seen in Close-Up, but Anti-Senescence in Long-Shot

SJR Q1Molecules and CellsOA

When mammalian cells and animals face a variety of internal or external stresses, they need to make homeostatic changes so as to cope with various stresses. To this end, mammalian cells are equipped with two critical stress responses, autophagy and cellular senescence. Autophagy and cellular senescence share a number of stimuli including telomere shortening, DNA damage, oncogenic stress and oxidative stress, suggesting their intimate relationship. Autophagy is originally thought to suppress cell

Senolytics and Senostatics: A Two-Pronged Approach to Target Cellular Senescence for Delaying Aging and Age-Related Diseases.
Chanhee Kang
PubMedOA

Aging is the most important single risk factor for many chronic diseases such as cancer, metabolic syndrome, and neurodegenerative disorders. Targeting aging itself might, therefore, be a better strategy than targeting each chronic disease individually for enhancing human health. Although much should be achieved for completely understanding the biological basis of aging, cellular senescence is now believed to mainly contribute to organismal aging via two independent, yet not mutually exclusive m

To be or not to be, the level of autophagy is the question: Dual roles of autophagy in the survival response to starvation
Chanhee Kang, Leon Avery
SJR Q1AutophagyOA

Autophagy is an evolutionally conserved lysosomal pathway used to degrade and turn over long-lived proteins and cytoplasmic organelles. Since autophagy was discovered, it has been thought to act as a pro-survival response to several stresses, especially starvation, at the cell and organism levels by providing recycled metabolic substrates to maintain energy homeostasis. However, several recent studies suggest that autophagy also plays a pro-death role through an autophagic cell death pathway mos

Systemic regulation of starvation response in Caenorhabditis elegans
Chanhee Kang, Leon Avery
SJR Q1Genes & DevelopmentOA

When the supply of environmental nutrients is limited, multicellular animals can make both physiological and behavioral changes so as to cope with nutrient starvation. Although physiological and behavioral effects of starvation are well known, the mechanisms by which animals sense starvation systemically remain elusive. Furthermore, what constituent of food is sensed and how it modulates starvation response is still poorly understood. In this study, we use a starvation-hypersensitive mutant to i

Process Design for the Sustainable Production of Butyric Acid Using Techno-Economic Analysis and Life Cycle Assessment

SJR Q1ACS Sustainable Chemistry & Engineering

Environmental pollution is gradually increasing owing to the increasing use of fossil fuels and fossil-derived chemicals. To overcome this issue, research on renewable and eco-friendly resources is being conducted. Biomass, which can replace fossil-based materials, has the advantages of high carbon efficiency and eco-friendliness. Therefore, we propose an integrated process for producing butyric acid, which is a precursor of biofuel, from white birch. Herein, cellulose is converted to butyric ac

SJR Q1FEBS JournalOA

Autophagy is an evolutionarily conserved lysosomal pathway used to degrade and recycle long-lived proteins and cytoplasmic organelles. This homeostatic ability makes autophagy an important pro-survival mechanism in response to several stresses, such as nutrient starvation, hypoxia, damaged mitochondria, protein aggregation and pathogens. However, several recent studies have highlighted that autophagy also acts as a pro-death mechanism. What on the surface seem like conflicting roles of autophagy

Dynamic changes in cell size are associated with development and pathological conditions, including aging. Although cell enlargement is a prominent morphological feature of cellular senescence, its functional implications are unknown; moreover, how senescent cells maintain their enlargement state is less understood. Here we show that an extensive remodeling of actin cytoskeleton is necessary for establishing senescence-associated cell enlargement and pro-inflammatory senescence-associated secret

Natural products and their derivatives historically represent alternatives to conventional synthetic molecules for pharmacotherapy, ranging from cancer chemotherapeutics to cosmetic ingredients that exert anti-aging activities. Cellular senescence is considered a main driver of skin aging, yet natural products that target skin senescence in a specific manner are not thoroughly explored. Here, we performed a focused compound screen to identify natural products that exert anti-senescence effects.

Systemic regulation of autophagy inCaenorhabditis elegans

SJR Q1AutophagyOA

When no supply of environmental nutrients is available, cells induce autophagy, thereby generating a source of emergency metabolic substrates and energy to maintain the basal cellular activity needed for survival. This autophagy response to starvation has been well characterized in various multicellular organisms, including worms, flies and mice. Although prosurvival effects of autophagy in response to starvation are well known in animals, the mechanisms by which animals regulate and coordinate
