연구실 소개
이창한 교수의 연구실은 단백질 공학과 약물 동역학을 기반으로 한 혁신적 치료제 개발에 초점을 맞추고 있습니다. 특히 항체 기반 약물의 체내 반감기를 연장시키는 Fc 도메인 변형 기술과, 암세포를 선택적으로 조절하는 크링글 도메인 기반 타겟 단백질 설계에 주력하고 있으며, RNA 간섭 기반의 정밀한 유전자 치료 전략 개발도 진행 중입니다. 이들의 연구는 암 치료 및 감염병에 대비한 고안된 생물학적 치료제의 설계 원리를 제시하고 있습니다.

Here, we report the development of target-specific binding proteins based on the kringle domain (KD) ( approximately 80 residues), a ubiquitous modular structural unit occurring across eukaryotic species. By exploiting the highly conserved backbone folding by core residues, but using extensive sequence variations in the seven loop regions of naturally occurring human KDs, we generated a synthetic KD library on the yeast cell surface by randomizing 45 residues in the loops of a human KD template.

The silencing of specific oncogenes via RNA interference (RNAi) holds great promise for the future of cancer therapy. Avulsion fracture of the acromion is rare. It is difficult to diagnosis because there is little displacement and it occurs even without direct trauma. - Addition of salt enhanced thermal stability of model substrate proteins by reducing electrostatic repulsion between protein molecules. - However, the opposite effect was observed with bacterial cell lysate, indicating that certain molecules within the lysate could enhance protein stability via electrostatic interactions. - Such molecules present in cell lysate were found to be nucleic acids known to have a potent anti-aggregation activity toward proteins involving electrostatic interactions. -
