연구실 소개

김학중 교수의 연구실은 주로 자연물 합성과 그 생합성 경로를 해독하는 데 초점을 맞추고 있으며, 특히 복잡한 구조를 가진 폴리케타이드 유도체인 스피노신 A와 아세티노박틴과 같은 철 흡착 색소의 생합성 및 기능 해석을 중심으로 연구를 진행하고 있습니다. 효소 기반의 총합성 전략과 생합성 유전자 클러스터 분석을 통해 천연물의 합성 경로를 규명하고, 이는 신약 타겟 발굴과 약물 개발에 기여하고자 합니다. 또한, 단백질 분해를 유도하는 새로운 약물 전략인 TPD(타겟 단백질 분해) 기반의 약물 개발 동향에 대해서도 종합적이고 전망적인 연구를 수행하고 있습니다.

The stereoselective synthesis of fimsbactin A, a siderophore of the human pathogen <i>Acinetobacter baumannii</i>, was established. Based on this synthetic route, various fimsbactin stereoisomeric analogues were generated and tested for their iron delivery activity for <i>A. baumannii</i>. This investigation revealed that the fimsbactin uptake machinery in this bacterium was indeed highly stereoselective in substrate recognition.

A novel fluorescence probe capable of assessing the cytoplasmic entry of siderophore-based conjugates was synthesized and evaluated by photochemical characterization and cell-based assays. The specific responsiveness to the cytoplasmic entry of the probe was implemented by adopting a disulfide linker, whose cleavage under the reducing conditions of the cytoplasm induced the display of a distinctive fluorescence signal.
