Therapy-related Acute Lymphoblastic Leukaemia has a Unique Genetic Profile Compared to De Novo Acute Lymphoblastic Leukaemia

<b>Background</b>: Unlike therapy-related myeloid neoplasms, therapy-related acute lymphoblastic leukaemia (tr-ALL) is poorly defined due to its rarity. However, increasing reports have demonstrated that tr-ALL is a distinct entity with adverse genetic features and clinical outcomes. <b>Methods:</b> We compared the clinicopathological characteristics and outcomes of patients diagnosed with tr-ALL (<i>n</i> = 9) or <i>de novo</i> ALL (dn-ALL; <i>n</i> = 162) at a single institution from January 2

Our highly sensitive and accurate NGS-MRD panel can be readily used to monitor most AML patients in clinical practice, including patients without gene rearrangement. In addition, this NGS-MRD panel may allow the detection of newly emerging clones during clinical relapse, leading to more reliable prognoses of AML.

BACKGROUND: A consolidation strategy has not been established for transplant-ineligible elderly patients with primary central nervous system lymphoma (PCNSL). In this study, we aimed to retrospectively evaluate the clinical outcomes of etoposide and cytarabine (EA) as consolidation chemotherapy for transplant-ineligible patients with PCNSL following high-dose methotrexate (MTX)-based induction chemotherapy. MATERIALS AND METHODS: Between 2015 and 2021, newly diagnosed transplant-ineligible patie

ABSTRACT: Emerging evidence indicates that CD4+ T cells contribute to antitumor immunity beyond their traditional roles as helpers or regulators. However, the specific subset of CD4+ T cells mediating beneficial outcomes in patients with multiple myeloma remains unclear. Here, we performed single-cell RNA sequencing and T-cell receptor sequencing on CD4+ T cells sorted from the bone marrow of patients across the stages of myeloma progression. We identified several distinct states of CD4+ cytotox

The demographics of Korean patients with POEMS syndrome were similar to those reported previously. Because of the introduction of new treatment agents and the reduced rate of transplant-related mortality related to ASCT, the treatment outcomes of Korean patients with POEMS syndrome have improved in recent years.

SJR Q2Blood ResearchOA

Abstract Purpose Allogeneic hematopoietic stem cell transplantation remains a curative option for acute leukemia. While an adequate CD34 + cell dose is essential for engraftment, the optimal upper threshold in haploidentical peripheral blood stem cell transplantation (haplo-PBSCT) remains unclear. Methods We retrospectively analyzed 81 patients with acute leukemia who underwent haplo-PBSCT with reduced-intensity conditioning between 2010 and 2020. Patients were stratified by CD34 + cell dose (&l

Background: Recent advancements in single-cell RNA sequencing have enhanced our understanding of the immunological dysfunctions during multiple myeloma (MM) progression from monoclonal gammopathy of undetermined significance (MGUS) and smoldering MM (SMM). Immunotherapies employing T cells have demonstrated promise in treating MM. However, a better understanding of effector T cells is necessary due to their limited response durability. While most forms of cancer immunotherapy have primarily focu

SJR Q2Blood ResearchOA

Abstract Purpose Allogeneic stem cell transplantation (allo-SCT) is a potentially curative treatment option for patients with relapsed or refractory lymphoid malignancies. However, the prognostic factors influencing survival outcomes in these patients remain poorly defined. This study aimed to evaluate the clinical variables associated with progression-free survival (PFS) and overall survival (OS) in patients undergoing allo-SCT for lymphoid malignancies. Methods We analyzed 58 patients who unde

Abstract Introduction:While the genetic landscape of acute lymphoblastic leukemia (ALL) at diagnosis has been deeply elucidated, its genetic evolution at relapse remains poorly understood. In this study, we compared the targeted next-generation sequencing (NGS) results of ALL before and after relapse and investigated their clinical significance. Methods:This was a retrospective study conducted at a single center, Severance hospital, in South Korea. Patients diagnosed with ALL who relapsed betwee
