Cellular heterogeneity of circulating CD4+CD8+ double-positive T cells characterized by single-cell RNA sequencing

SJR Q1Scientific ReportsOA

Abstract Circulating CD4 + CD8 + double-positive (DP) T cells are associated with a variety of disease states. However, unlike conventional T cells, the composition of this population is poorly understood. Here, we used single-cell RNA sequencing (scRNA-seq) to analyze the composition and characteristics of the DP T cell population circulating in the peripheral blood of cynomolgus monkeys. We found that circulating DP T cells not only contain a large number of naïve cells, but also comprise a he

Kübler Illite “Crystallinity” Index of the Cretaceous Gyeongsang Basin, Korea: Implications for Basin Evolution

SJR Q2Clays and Clay Minerals

Abstract Thermal maturity of the Lower Cretaceous Sindong and Hayang groups in the Gyeongsang Basin, Korea, was investigated using the Kübler illite “crystallinity” index (KI) which is based on the numerical expression of the 10-Å peak width after calibration to the Crystallinity Index Standard scale. The metamorphic grade of the Sindong and Hayang groups ranges from late diagenetic zone to high anchizone. Depth of burial was not a major factor controlling KI variation in the basin because Hayan

Prognostic Effects of Adjuvant Chemotherapy-Induced Amenorrhea and Subsequent Resumption of Menstruation for Premenopausal Breast Cancer Patients

SJR Q3MedicineOA

Chemotherapy-induced amenorrhea (CIA) is a side effect that occurs in patients with breast cancer (BC) as a result of chemotherapy. These patients require special treatments to avoid infertility and menopause. However, the factors controlling CIA, resumption of menstruation (RM), and persistence of menstruation after chemotherapy are unknown. The long-term prognosis for premenopausal patients with BC and the prognostic factors associated with CIA and RM are subject to debate. We performed a retr

CD4hiCD8lowDouble-Positive T Cells Are Associated with Graft Rejection in a Nonhuman Primate Model of Islet Transplantation
Yun Jung Choi, Hi-Jung Park, Hyejin Park, Kyeong Cheon Jung, Jae Il Lee
SJR Q1Journal of Immunology ResearchOA

Peripheral CD4/CD8 double-positive (DP) T cells are associated with autoimmune disorders, cancer, and viral infection. However, the relationship between organ transplantation and DP T cells is unclear. Here, we examined the functional characteristics of peripheral DP T cells and analyzed their significance with respect to islet graft rejection in a nonhuman primate model of islet transplantation. DP T cells were functionally equivalent to conventional CD4 and CD8 T cells in terms of helper and c

Abstract The frequency of CD4 + CD8 + double-positive (DP) T cells is highly associated with a variety of diseases. Recently, we used high-throughput single-cell RNA sequencing to show that circulating DP T cells in cynomolgus monkeys comprise nine heterogeneous populations. To better understand the characteristics of DP T cells, we analyzed 7601 cells from a rhesus monkey and detected 14,459 genes. Rhesus monkey DP T cells comprised heterogeneous populations (naïve, Treg-, Tfh-, CCR9 + Th-, Th1

TaqMan Probe Real-Time PCR for Quantitative Detection of Mycoplasma during Manufacture of Biologics

KSBB JournalOA

Mycoplasma is well recognized as one of the most prevalent and serious microbial contaminants of biologic manufacturing processes. Conventional methods for mycoplasma testing, direct culture method and indirect indicator cell culture method, are lengthy, costly and less sensitive to noncultivable species. In this report, we describe a new TaqMan probe-based real-time PCR method for rapid and quantitative detection of mycoplasma contamination during manufacture of biologics. Universal mycoplasma

IL-4/IL-4 Ab complex enhances the accumulation of both antigen-specific and bystander CD8 T cells in mouse lungs infected with influenza A virus
Hi Jung Park, Eun A Choi, Sung Min Choi, Young Ki Choi, Jae Il Lee, Kyeong Cheon Jung
SJR Q2Laboratory Animal ResearchOA

BACKGROUND: Unlike conventional T cells, innate and virtual-memory CD8 T cells in naïve mice acquire their memory phenotypes and functions in the absence of antigenic encounters in a cytokine-dependent manner. The relevant cytokines include interleukin-4 (IL-4), type I interferon, and interleukin-15 (IL-15). Moreover, exogenous IL-4 can also induce de novo generation and/or expansion of the virtual-memory CD8 T cell population. In this study, we investigated whether exogenous IL-4 could enhance

Characterization of CD8+ virtual memory T cells in IL-4 knockout mice using single-cell RNA sequencing

SJR Q2Biochemical and Biophysical Research CommunicationsOA

Antigen-inexperienced memory-phenotype CD8+ T cells are categorized as innate memory cells in the thymus or virtual memory (VM) CD8+ T cells in peripheral tissues. The key distinction between these cell types is their differing responses to IL-4, but the minimal effect of IL-4 on VM CD8+ T cell expansion in the periphery is not well understood. To address this, we investigated the development of VM CD8+ T cells in the periphery of IL-4 knockout (KO) C5

Kinetics of an alternating copolymer model

Physical review. A, General physics

The time-dependent behavior of a model spin system which might represent certain properties of alternating copolymers is studied. This model is an extension of the kinetic Ising model of Glauber to a system in which the strengths of interactions between neighboring spins alternate. The exact stationary and time-dependent solutions for the average spin and spin-pair correlations are obtained for this system. It exhibits two relaxation times, in contrast to the Glauber model; this feature may be r

IL‐4, IL‐15, and Type I Interferon Orchestrate the Shaping of the Heterogeneity of Virtual Memory CD8 T Cells

SJR Q1European Journal of ImmunologyOA

ABSTRACT The development of virtual memory CD8 T cells is dependent on IL‐4, type I interferon, and IL‐15. However, it remains unclear whether these cytokines individually contribute to the generation of specific subsets of virtual memory CD8 T cells. In this study, virtual memory CD8 T cells were categorized into four subsets based on Ly6C and Sca‐1 expression, and their development was examined using knock‐out mice lacking IFNAR1, IL‐4, or IL‐15Rα. Notably, both Ly6C + Sca‐1 + and Ly6C − Sca‐1

SJR Q2Laboratory Animal ResearchOA

BACKGROUND: Influenza A viruses (IAVs) have long posed a threat to humans, occasionally causing significant morbidity and mortality. The initial immune response is triggered by infected epithelial cells, alveolar macrophages and dendritic cells. However, an exaggerated innate immune response can result in severe lung injury and even host mortality. One notable pathology observed in hosts succumbing to severe influenza is the excessive influx of neutrophils and monocytes into the lung. In this st

CD1b glycoprotein, a crucial marker of thymocyte development during T cell maturation in cynomolgus monkeys
Sung Min Choi, Hi Jung Park, Eun A Choi, Kyeong Cheon Jung, Jae Il Lee
SJR Q1Scientific ReportsOA

Abstract Phenotypic markers that denote different developmental stages of thymocytes are important for understanding T cell development in the thymus. Here, we show that CD1b is a critical discriminator of thymocyte maturation stage in cynomolgus monkeys. CD1b was expressed by immature thymocytes prior to β-selection, and its expression decreased as cells became fully mature in the thymus. MHC-I expression was lowest at the CD3 lo CD1b + immature double-positive (DP) stage, while the ratio of CD
