연구실 소개

안정혁 교수의 연구실은 난소세포암의 전이 메커니즘을 밝히기 위해 유전자 발현, 에피제네틱 조절, 단백질 인산화 조절 등 다각도의 분자 생물학적 접근을 펼치고 있습니다. 특히 GABRP, MUC13, CA9와 같은 전이 관련 유전자 및 PP2A를 중심으로 한 타우 단백질 인산화 조절 메커니즘을 규명하며 암의 치료 저항성과 전이의 분자 기반을 탐색하고 있습니다. 알츠하이머병 관련 유전자(APP-swe)의 전사체 분석과 함께 신경퇴행성 질환에서도 분자 조절 네트워크의 기초를 다지고 있습니다.

Aberrant epigenetic regulation of GABRP associates with aggressive phenotype of ovarian cancer

Metastasis is a major cause of therapeutic failure in ovarian cancer. To elucidate molecular mechanisms of ovarian cancer metastasis, we previously established a metastatic xenograft mouse model using human ovarian carcinoma SK-OV-3 cells. Phosphorylation on the PPP2R5D B regulatory subunit modulates the biochemical properties of protein phosphatase 2A

To characterize the biochemical properties of the PP2A regulatory B subunit, PPP2R5D, we analyzed its phosphorylation sites, stoichiometry and effect on holoenzyme activity. PPP2R5D was phosphorylated on Ser-53, Ser-68, Ser-81, and Ser-566 by protein kinase A, and mutations at all four of these sites abolished any significant phosphorylation in vitro.
