Microscale electroporation: challenges and perspectives for clinical applications
Won Gu Lee, Utkan Demirci, Ali Khademhosseini
SJR Q3Integrative BiologyOA

Microscale engineering plays a significant role in developing tools for biological applications by miniaturizing devices and providing controllable microenvironments for in vitro cell research. Miniaturized devices offer numerous benefits in comparison to their macroscale counterparts, such as lower use of expensive reagents, biomimetic environments, and the ability to manipulate single cells. Microscale electroporation is one of the main beneficiaries of microscale engineering as it provides sp

Electroporation for nanomedicine: a review
Ki-Soo Kim, Won Gu Lee
SJR Q1Journal of Materials Chemistry B

Nanoparticles have shown great promise in the development of efficient drug delivery systems, early diagnosis, and high-resolution imaging of hard-to-find diseases, such as early-stage malignant cancer and very rare circulating tumor cells, for pharmaceutical and medical applications. Recently, nanoparticles have been used as intracellular carriers of nano-drugs into targeted cells to release a slowly diffusing drug in the vicinity of the target disease. Several methods such as chemical reagent-

A Hollow Sphere Soft Lithography Approach for Long-Term Hanging Drop Methods
Won Gu Lee, Daniel Ortmann, Matthew Hancock, Hojae Bae, Ali Khademhosseini
SJR Q2Tissue Engineering Part C MethodsOA

In conventional hanging drop (HD) methods, embryonic stem cell aggregates or embryoid bodies (EBs) are often maintained in small inverted droplets. Gravity limits the volumes of these droplets to less than 50 microL, and hence such cell cultures can only be sustained for a few days without frequent media changes. Here we present a new approach to performing long-term HD methods (10-15 days) that can provide larger media reservoirs in a HD format to maintain more consistent culture media conditio

Wearable sensors for continuous oral cavity and dietary monitoring toward personalized healthcare and digital medicine
Wonki Hong, Won Gu Lee
SJR Q2The Analyst

Oral monitoring plays an essential role in preventing and diagnosing systemic diseases through saliva in the mouth. Dietary monitoring is also crucial to reduce the likelihood of chronic diseases such as hypertension and diabetes by analyzing food types, amounts and diet patterns. Therefore, the oral cavity and dietary monitoring are vital for accurate personalized healthcare management that can improve healthcare. To perform continuous oral cavity and dietary monitoring in real-time, a wearable

Continuous Ethanol Production from Concentrated Wood Hydrolysates in an Internal Membrane-Filtration Bioreactor
Won Gu Lee, Borae G. Park, Yong Keun Chang, H. CHANG, Jung Soo Lee, Su-Beom Park
SJR Q2Biotechnology Progress

Continuous culture for the production of ethanol from wood hydrolysate was carried out in an internal membrane-filtration bioreactor. The hydrolysate medium was sterilized at a relatively low temperature of 60 degrees C with the intention of reducing the formation of inhibitory compounds during the sterilization. The maximum ethanol concentration and productivity obtained in this study were 76.9 g/L and 16.9 g/L-h, respectively, which were much higher than those (57.2-67 g/L and 0.3-1.0 g/L-h) o

SJR Q1Lab on a Chip

This paper presents a novel method for an on-chip erythrocyte deformability test under optical pressure, especially to enhance the level of sensitivity with respect to the detection of cancerous diseases. To demonstrate the performance and sensitivity of the combined method, we introduce the concept of transit velocity, a modified elongation index, and shape recovery time of individual erythrocytes in a strictly confined region (2 microm deep, 4 microm wide, and 100 microm long). Finally, we inv

Microfabricated particulate drug‐delivery systems
Jing Pan, Sui Yung Chan, Won Gu Lee, Lifeng Kang
SJR Q2Biotechnology Journal

Micro- and nanoparticulate drug-delivery systems (DDSs) play a significant role in formulation sciences. Most particulate DDSs are scaffold-free, although some particles are encapsulated inside other biomaterials for controlled release. Despite rapid progress in recent years, challenges still remain in controlling the homogenicity of micro-/nanoparticles, especially for two crucial factors in particulate DDSs: the size and shape of the particles. Recent approaches make use of microfabrication te

Seeing the electroporative uptake of cell-membrane impermeable fluorescent molecules and nanoparticles
Ki-Soo Kim, Jeong Ah Kim, Soon‐Geul Lee, Won Gu Lee
SJR Q1Nanoscale

This paper presents direct visualization of uptake directionality for cell-membrane impermeant fluorescent molecules and fluorescence-doped nanoparticles at a single-cell level during electroporation. To observe directly the uptake direction, we used microchannel-type electroporation that can generate a relatively symmetric and uniform electric field. For all the image frames during electroporation, fluorescence intensities that occurred at cell membranes in both uptake directions toward the ele

Role of weakly polarized nanoparticles in electroporation
Jeong Ah Kim, Won Gu Lee
SJR Q1Nanoscale

In this study, we present a role for weakly polarized nanoparticles as engineered gene transporters that can enhance electromediated gene delivery. To validate this proof-of-concept, fluorescent poly(ethylene glycol) (PEG)-coated silica nanoparticles (SiNPs) with opposite polarities, SiNPs(RITC)-PEG/PTMA(+) and SiNPs(RITC)-PEG/PMP(-), are used. To investigate the electroporative uptake direction of the nanoparticles-gene complex, we employ microscale electroporation to generate more symmetric an
