In order for engineered tissues to find clinical utility, the engineered tissues must function appropriately. However, smooth muscle (SM) tissues engineered in vitro with a conventional tissue engineering technique may not exhibit contractile functions, because smooth muscle cells (SMCs) cultured in vitro typically revert from a contractile, differentiated phenotype to a synthetic, nondifferentiated phenotype and lose their ability to contract. SMCs in vivo typically reside in mechanically dynam

Nonviral delivery for reprogramming to pluripotency and differentiation
박현지, 신지수, 김진, 조승우

Nonviral delivery is a promising strategy forcellular reprogramming to produce desired cell types fromundifferentiated stem cells or terminally differentiatedsomatic cells. Nonviral delivery of genes (DNA, RNA),proteins, or peptides has the potential to reprogram somaticcells to pluripotent stem cells or other lineage cells, and topromote the differentiation of stem cells to specific lineages. Various delivery carriers (cationic polymers, lipids, scaffolds, transposons, cell-penetrating peptides)

Hydrogel-integrated Microfluidic Systems for Advanced Stem Cell Engineering
안수환, 한승엽, 조승우

Previous culture techniques are often ineffective for providing appropriate conditions to cells grown in vitro for efficient growth and maturation. However, the advent of microfluidic chips allows us to manipulate various factors from co-culturing cells to inducing shear stress and biochemical gradient. The above have all been effectively applied to stem cell engineering, allowing dynamic interactions with other cells and, as a result, acquisition of a more mature state. The introduction of both

Cell type specification is a delicate biological event in which every step is under tight regulation. From a molecular point of view, cell fate commitment begins with chromatin alteration, which kickstarts lineage-determining factors to initiate a series of genes required for cell specification. Several important neuronal differentiation factors have been identified from ectopic over-expression studies. However, there is scarce information on which DNA regions are modified during induced pluripo

Liver Tissue Engineering: Recent Advances in the Development of a Bio-artificial Liver
Jung Seung Lee, 조승우

Orthotopic liver transplantation is the most common treatment for patients with end-stage liver failure. However, liver transplantation is greatly limited by a donor shortage. Liver tissue engineering may offer a promising strategy to solve this problem by providing transplantable, bioartificial livers. Diverse types of cells, biomaterials, and growth factor delivery systems have been tested for efficient regeneration of liver tissues that possess hepatic functions comparable to native livers. Th

Implantable Microfluidic Device for the Formation of Three-dimensional Vasculature by Human Endothelial Progenitor Cells
김진, 양기석, 박현지, 한세운, 신유진, 이준협, 정석, 조승우

Vasculogenesis is an important morphogeneticevent for vascular tissue engineering and ischemic diseasetreatment. Stem and progenitor cells can contribute tovasculogenesis via endothelial differentiation and directparticipation in blood vessel formation. In this study, wedeveloped an implantable microfluidic device to facilitateformation of three-dimensional (3D) vascular structures byhuman endothelial progenitor cells (hEPCs). The microfluidicdevice was made of biodegradable poly(lactic-co-glyco

Diabetes and Metabolism Journal

Over the past three decades, human pancreatic islet isolation and transplantation techniques have developed as a routine clinical procedure for selected patients with type 1 diabetes mellitus. However, due to the donor shortage and required chronic systemic immunosuppression, the widespread application of islet transplantation is limited. To overcome these limitations, providing a physical barrier to transplanted islet cells with encapsulating biomaterial has emerged as a promising approach to e

Gastrointestinal tract modeling using organoids engineered with cellular and microbiota niches
민성진, Suran Kim, 조승우

The recent emergence of organoid technology has attracted great attention in gastroenterology because the gastrointestinal (GI) tract can be recapitulated in vitro using organoids, enabling disease modeling and mechanistic studies. However, to more precisely emulate the GI microenvironment in vivo, several neighboring cell types and types of microbiota need to be integrated into GI organoids. This article reviews the recent progress made in elucidating the crosstalk between GI organoids and comp

Endothelial-neurosphere crosstalk in microwell arrays regulates self-renewal and differentiation of human neural stem cells

Control of neural stem cell (NSC) self-renewal and differentiationis of great importance to improve itstherapeutic efficacy in the treatment of neurodegenerative diseases. Neurosphere culture for NSCexpansion under undifferentiation condition determines the self-renewal capacity and differentiationpropensity of NSCs. In this study, we examined the effects of controlled crosstalk between endothelialcells (ECs) and NSC neurospheres on self-renewal, differentiation, and functions of NSCs. Cultures

In the present study, we tested the hypothesis that granulocyte colony-stimulating factor (G-CSF) administration would enhance the efficacy of cellular cardiomyoplasty with embryonic stem (ES) cells in infarcted myocardium. Three weeks after myocardial infarction by cryoinjury, Sprague-Dawley rats were randomized to receive either an injection of medium, ES cell transplantation, G-CSF administration, or a combination of G-CSF administration and ES cell transplantation. The purpose of this study was to identify the angiogenesis effect by transplantion of the autologous whole bone marrow stem cells(BMSCs) and bone marrow derived mononuclear cells(BM-MNCs). A chronic ischemic hind limb model was created by placing an ameroid constrictor around a femoral artery with in a dog model. Animals were divided into two groups. In the BMSC group, about 20 ml of BMSCs were aspirated from the femur and then injected into the medial side of the affected thigh muscle, and in t

SJR Q2Journal of Microbiology and BiotechnologyOA

The extracellular matrix (ECM) plays a central role in directing dermal fibroblast behavior and coordinating tissue regeneration through its structural organization and biochemical signaling. In this study, we investigate the composition and regenerative bioactivity of Skin ECM in the context of dermal remodeling as a soluble supplement and surface coating for fibroblast culture. Proteomic profiling demonstrates that Skin ECM preserves the molecular complexity and skin-specific composition of na
