연구실 소개

이 교수의 연구실은 고도로 분해된 생체 샘플, 특히 오래된 뼈 및 유골에서의 DNA 회수와 분석 기술 개발에 초점을 맞추고 있습니다. 특히, 미세한 DNA 손실을 최소화하고 불순물 영향을 줄이기 위한 고처리량 시퀀싱(МPS) 기반 STR 분석 및 이온 교환 컬럼을 활용한 정제 기법을 개발하고 있습니다. 또한, 유사한 유전적 배경을 가진 일란성 쌍둥이를 구별할 수 있는 DNA 메틸로믹스 분석을 통해 법의학적 정황 해석의 정밀도를 높이는 데에도 기여하고 있습니다.

We investigated whether a combination of recently introduced methods, total demineralization and ion-exchange columns, would increase DNA recovery from old bone. Ten bone samples taken after a burial period of approximately 60 years were used in this study. Bone powder was digested using total or incomplete demineralization. The effects on STR typing results, using a two-step PCR protocol that combines the annealing and extension steps, were determined using samples at various DNA concentrations. BACKGROUND: Short tandem repeat (STR) markers cannot be used to distinguish between genetically identical monozygotic (MZ) twins, causing problems in a case with an MZ twin as a suspect. Many studies have shown that in older MZ twins, there are significant differences in overall content and genomic distribution of methylation. OBJECTIVE: In this study, we analyzed the DNA methylome profile of blood to identify recurrent differentially methylated CpG sites (DMCs) to discriminate between MZ twins.
