15The visible light-cured glycol chitosan (GC) hydrogel systems for the sustained release of growth factors (GFs) were prepared, and their efficacies in wound healing acceleration were investigated. Vascular endothelial growth factor (VEGF) and platelet-derived growth factor-BB (PDGF-BB) were selected as the dual GFs and commercially available Duoderm1 was used as control. In order to increase intermolecular chain mobility, methoxy (polyethylene glycol) acetic acid (MPEG-COOH) was conjugated to GC
Background Recently, various studies have revealed that 3D cell spheroids have several advantages over 2D cells in stem cell culture. However, conventional 3D spheroid culture methods have some disadvantages and limitations such as time required for spheroid formation and complexity of the experimental process. Here, we used acoustic levitation as cell culture platform to overcome the limitation of conventional 3D culture methods. Methods In our anti-gravity bioreactor, continuous standing sonic
We report a facile one-pot aqueous-phase synthesis of PdAu bimetallic nanoparticles with different Pd/Au ratio. The synthesis was conducted by co-reduction of Pd and Au precursor using ascorbic acid as a reducing agent and in the presence of polyallylamine hydrochloride (PAH). By high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM) and energy-dispersive X-ray spectrometry (EDS) analyses, we found that the synthesized nanoparticles had an onion-like core/shell/shel
This work describes a simple aqueous-phase synthesis of single-crystal ZnO nanobolts with sizes of250 nm by reacting Zn(NO3)2 with octylamine. The synthesized ZnO nanobolts exhibit a hierarchicalstructure consisting of two well-defined hexagonal nanorods of different sizes. In a study of the growthbehavior of the ZnO nanobolts, we observed that aggregates of around 600 nm in size became single-crystal ZnO nanobolts with sizes of 250 nm. We also investigated the influence of reaction temperaturea
Due to their heat-generating property based on exogenous energy irradiation, reduced graphene oxideparticles (RGOs) have been widely used for photothermal cancer therapy. They show high absorbance forthe near-infrared wavelength that is suitable for in vivo experimentation. However, enhancing the tumor-targeting efficiency and avoiding the self-aggregation of RGO to maximize the photothermal effectremain challenging. We demonstrated here that human mesenchymal stem cells (hMSCs) can deliverpolox
Transplantation of stem cell spheroids to ischemic tissues has been suggested as an effective method fortreating ischemic disease, but its therapeutic efficacy varies according to the preparation methods forspheroids. We hypothesized that increasing cell compaction while preparing spheroids could improvethe angiogenic efficacy of spheroids. Optimized human adipose derived stem cell (hADSC) spheroidswere shown to upregulate cell adhesion molecule, and angiogenic paracrine factors expression withd
BACKGROUND: Stem cell-based therapies have been developed to treat various types of wounds. Human adiposederived stem cells (hADSCs) are used to treat skin wounds owing to their outstanding angiogenic potential. Although recent studies have suggested that stem cell spheroids may help wound healing, their cell viability and retention rate in the wound area require improvement to enhance their therapeutic efficacy. METHODS: We developed a core–shell structured spheroid with hADSCs in the core and
Cell-based therapies have been used as promising treatments for several untreatable diseases. However, cell-based therapies have side effects such as tumorigenesis and immune responses. To overcome these side effects, therapeutic effects of exosomes have been researched as replacements for cell-based therapies. In addition, exosomes reduced the risk that can be induced by cell-based therapies. Exosomes contain biomolecules such as proteins, lipids, and nucleic acids that play an essential role i
BACKGROUND: Enhancing blood flow and cell proliferation in the hair dermis is critical for treating hair loss. This study was designed to aid the development of alternative and effective solutions to overcome alopecia. Specifically, we examined the effects of Morus alba. L root extract (MARE, which has been used in traditional medicine as a stimulant for hair proliferation) on dermal fibroblasts and other cell types found in the epidermis. METHODS: We first optimized the concentration of MARE th
BACKGROUND: Owing to the tumor-targeted migration capacity of human mesenchymal stem cells (hMSCs), they have been combined with nanoparticles for photothermal therapy. However, the low viability of hMSCs following transplantation remains a problem. Here, we developed iron (Fe) ion-releasing gold (Au) nanoparticles (IIAuNPs) for advanced tumor-targeted photothermal therapy using hMSCs. METHODS: IIAuNPs were designed to undergo degradation under low pH conditions, such as the endosomal microenvir
BACKGROUND: Various methods based on gold nanoparticles (AuNPs) have been applied to enhance the photothermal effect. Among these methods, combining gold nanoparticles and stem cells has been suggested as a new technique for elevating the efficiency of photothermal therapy (PT) in terms of enhancing tumor targeting effect. However, to elicit the efficiency of PT using gold nanoparticles and stem cells, delivering large amounts of AuNPs into stem cells without loss should be considered. METHODS:
A polyspermine-derived bioreducible polymer was synthesized as efficient siRNA carriers. Spermine was polymerized using disulfide-bearing cross-linkers, resulting in poly(disulfide spermine) (PDS). PDS was also conjugated with poly(ethylene glycol) (PEG) to tailor siRNA condensation efficiency of the polymer. PEG-conjugated PDS (PEG-PDS) was able to condense siRNA into nano-sized polyplexes at high N/P ratios. siRNA condensation efficiency of PDS was reduced by PEGylation due to its weakened int
Curcumin is a polyphenol extracted from the roots of Curcuma plants that exerts potential anticancer effects. However, owing to its low toxicity, curcumin is known to be effective only at high doses. Here, we report the highly sensitive assessment of curcumin toxicity in human liver cancer cells, which was facilitated using a fabricated conductive platform and an electrochemical detection method. To identify the best platform for assessing cell viability, both electrochemical deposition time (0–
Myocardial infarction (MI) is treated with stem cell transplantation using various biomaterials and methods, such as stem cell/spheroid injections, cell sheets, and cardiac patches. However, current treatment methods have some limitations, including low stem cell engraftment and poor therapeutic effects. Furthermore, these methods cause secondary damage to heart due to injection and suturing to immobilize them in the heart, inducing side effects. In this study, we developed stem cell spheroid-la
