연구실 소개

김선창 교수의 연구실은 주로 병원성 미생물을 타겟으로 하는 천연 항미생물 펩타이드의 분리 및 기능 해석을 핵심으로 삼고 있습니다. 특히 고등생물, 특히 어류와 양서류에서 유래한 펩타이드의 구조-기능 관계를 규명하고, 이들의 면역 방어 기전을 분자생물학적·생화학적 방법으로 규명하고 있습니다. 또한 항생제 내성 문제를 해결하기 위한 새로운 약물 개발을 위한 설계 원리를 제시하는 합성 생물학적 접근도 함께 수행하고 있습니다.

Parasin I, an antimicrobial peptide derived from histone H2A in the catfish, Parasilurus asotus

In response to epidermal injury, Parasilurus asotus, a catfish, secreted a strong antimicrobial peptide into the epithelial mucosal layer. The molecular mass of the antimicrobial peptide, named parasin I, was 2000.4 Da, as determined by matrix-associated laser desorption ionization mass spectrometry. A novel antimicrobial peptide, named misgurin, was isolated and characterized from the loach (mudfish), Misgurnus anguillicaudatus. The 21-amino-acid peptide with a molecular mass of 2502 Da was purified to homogeneity using a heparin-affinity column and C18 reverse-phase and gel-permeation high-performance liquid chromatography. Salt sensitivity of antimicrobial peptides poses a major obstacle in their development as novel antibiotics.
