연구실 소개

박윤규 교수의 연구실은 전사 조절 단백질을 표적으로 삼아 유전자 발현을 정밀하게 조절하는 신약 개발 전략을 핵심으로 삼고 있습니다. 특히, 전사 인자와의 상호작용을 방해하는 '도우미 올리고뉴클레오티드(Decoy Oligonucleotide)' 기반 치료 기법과 cAMP-PKA 신호전달 경로의 암 발생 관련 기전을 규명하는 데 주력하고 있습니다. 또한, 암세포에서 특이적으로 분비되는 단백질을 바이오마커로 활용하는 조기 진단 기술 개발도 함께 진행 중입니다. 연구는 암 치료와 조기 진단의 새로운 전략을 모색하는 데 초점을 맞추고 있습니다.

Overexpression of cAMP-dependent protein kinase (PKA) type I isozyme is associated with cell proliferation and neoplastic transformation. The presence of PKA on the external surface of LS-174T human colon carcinoma cells has been shown. Here, we show that cancer cells of various cell types excrete PKA into the conditioned medium. This extracellular PKA (ECPKA) is present in active, free catalytic subunit (C subunit) form, and its activity is specifically inhibited by PKA inhibitory protein, PKI.

Taken together, our results demonstrate that both replication and gene expression of HBV require a functional CREB and HBV-CRE. We have also demonstrated that CRE decoy oligonucleotides and the overexpression of CREB mutants can effectively block the HBV life cycle, suggesting that interventions against CREB activity could provide a new avenue to treat HBV infection.
